A major advance in environmental DNA (eDNA) metabarcoding has been announced with the launch of MitoNGS, a next-generation platform designed for high-resolution analysis of fish biodiversity. This new tool directly addresses critical challenges in evolutionary biology and population genetics, such as incomplete reference databases and ambiguous taxa sharing identical DNA barcodes, which have hindered accurate species-level identification. MitoNGS incorporates more comprehensive genetic references, including non-fish species, and introduces an innovative “species complex” strategy that integrates environmental habitat and geographic occurrence data to resolve taxonomic uncertainties. Furthermore, it expands analytical capabilities beyond its predecessor, the MiFish pipeline, by supporting data from any mitochondrial marker and from emerging Nanopore sequencing platforms, demonstrating excellent performance across diverse testing datasets.
