Investigators identified a novel microRNA-based regulatory mechanism in major depressive disorder (MDD), showing that miR-20a-5p directly targets the IRF1 3′-UTR to suppress its expression. In postmortem hippocampal tissue from MDD patients and in chronic social defeat stress mice, both IRF1 mRNA and miR-20a-5p were significantly upregulated, with functional assays confirming direct binding and early stress-mediated upregulation of Irf1 preceding secondary miRNA modulation. This study adds to the molecular understanding of neuroinflammation in MDD, offering a specific axis that could serve as a target for future blood-based diagnostic or therapeutic monitoring strategies, though the authors emphasize that in vivo functional significance remains to be established.
