A large-scale genomic study has identified a distinct genetic signature in children with ADHD who also present with comorbid neurodevelopmental disorders (NDDs) such as autism spectrum disorder and anxiety. By analyzing copy number variant (CNV) data from over 72,000 pediatric participants, researchers found significant enrichment of CNVs within glutamatergic signaling networks involving genes that interact with metabotropic glutamate receptors (GRMs). This work reveals that disruptions in GRM-interacting pathways, including key synaptic genes like DLG2, NRXN1, SHANK3, and SYNGAP1, are specifically associated with the more complex clinical profiles seen in comorbid ADHD, offering a potential avenue for genetically informed interventions.
