A population pharmacokinetic/pharmacodynamic model using 5,918 plasma samples from 743 patients established MTX concentration thresholds for nephrotoxicity risk in primary central nervous system lymphoma treatment: 9.71, 0.81, and 0.26 μmol/L at 24, 48, and 72 hours. This provides a practical tool for early identification and monitoring of methotrexate-induced renal injury.
