CRISPR-Cas9-generated rhbdl2 mutants in zebrafish reveal that the intramembrane serine protease Rhbdl2 acts as a restraint on immune-driven wound repair, with its loss producing accelerated tissue regeneration after injury. Mechanistically, rhbdl2 loss elevated Rac2 protein levels, increased macrophage migration speed and accumulation at the wound site, and was accompanied by heightened early apoptosis and cell proliferation; morpholino-mediated Rac2 knockdown suppressed both the enhanced macrophage recruitment and the accelerated repair phenotype. For research into fetal programming and the cellular mechanisms underlying fertility and ovarian aging, this work identifies a protease-controlled checkpoint that couples apoptotic signalling, cell proliferation, and innate immune cell trafficking — precisely the axis through which intrauterine and environmental exposures are thought to durably reshape tissue homeostasis and reproductive outcomes across the lifespan.
