This study reveals that the lipid kinase PIKfyve governs endoplasmic reticulum (ER) architecture and dynamics by modulating phosphoinositide balance at ER–lysosome contact sites. The researchers demonstrated that PIKfyve inhibition causes lysosomes to cluster perinuclearly and become less motile, arresting an ER “hitchhiking” process in which lysosomes pull and form ER tubules, while also promoting hyper-tethering of the ER protein protrudin to lysosomes via excess PtdIns(3)P.
