The Molecular Link Between Sleepless Nights and Alzheimer’s Pathology
A new study in Alzheimer’s & Dementia provides compelling longitudinal evidence that sleep disturbance is not just a symptom but a potential driver of Alzheimer’s disease pathology. Analyzing over 1,200 participants from the Alzheimer’s Disease Neuroimaging Initiative, researchers found that baseline sleep disturbance was associated with a significantly faster rate of decline in cerebrospinal fluid amyloid-beta 42 levels—a key biomarker of Alzheimer’s progression. To uncover the molecular mechanisms, the team integrated CSF proteomics with weighted gene co-expression network analysis, identifying a specific protein module that connects sleep disruption to Aβ dynamics.
